چکیده
A comparison of spermatozoa from intact boars and from boars without seminal vesicles, frozen in various diluents, was made to determine any differences in sperm quality after thawing and the conception rates of gilts. Sperm samples were frozen at -196 degrees C. 50 semen pellets were thawed after 1-3 weeks storage and assessed for postthaw motility at 37 degrees C. The percentage of eosinophilic spermatozoa was estimated. Random samples were assayed for L-aspartate 2-oxoglutarate aminotransferase activity. Enzyme release was measured. Gilts were inseminated 24 and 32 hours after 1st standing to a boar and gave slightly higher conception rates than those from intact animals although the differences were insignificant statistically. Overall conception rate was 35% for spermatozoa from boars without seminal vesicles and 29% for those who were intact. The diluents with 20% egg yolk and 2% casin with cholesterol and phosphatidyl serine provided significantly greater (p less than .05) cryoprotection than did that with 2% casein. In general, levels of enzyme release from the spermatozoa after thawing were 3 times those before freezing, indicating substantial membrane damage. It is suggested that the presence of seminal vesicular fluid at ejaculation has only a small effect on the fertilizing ability of spermatozoa frozen and thawed in suitable diluents.

P-65: Low-Density Lipoproteins Extractions on Boar Spermatozoa Quality Following Freezing–Thawing
Background Low-density lipoproteins (LDL) is known to protect boar sperm during freezing–thawing, but little information is known about the effects of LDL extracted from different avian egg yolks on post-thaw boar semen quality. The purpose of this study was to compare and analyze the effects of LDL at various concentrations and different species on boar sperm quality after freezing–thawing. Ma...
متن کاملCryosurvival and in vitro fertilizing capacity postthaw is improved when boar spermatozoa are frozen in the presence of seminal plasma from good freezer boars.
The study evaluated the protective effect of seminal plasma (SP) added to freezing extender against cryopreservation injuries to boar spermatozoa. Pooled sperm-rich fractions collected from 9 fertile boars were frozen in 0.5-mL straws after being extended in a conventional freezing extender either alone or supplemented with 5% of SPs (SP1-SP4) collected from the sperm-rich fractions (diluted 1:...
متن کاملApplication of seminal plasma in sex-sorting and sperm cryopreservation.
Substantial dilution of boar semen during processing decreased the concentration of seminal plasma, perhaps contributing to the decline in sperm quality after cryopreservation and sex-sorting. Results of replacing seminal plasma in investigations from many laboratories have been contradictory. Results and discussion here suggest that whereas membrane status can be influenced by seminal plasma, ...
متن کاملInfluence of seminal plasma on the kinematics of boar spermatozoa during freezing.
Sperm motility is, for its relation to cell viability and fertility, a central component of the spermiogram, where consideration of motion patterns allows discrimination of sub-populations among boar spermatozoa. Extension and cryo-preservation imposes changes in these patterns in connection to handling, additives, temperature changes and the removal of boar seminal plasma (BSP) which apparentl...
متن کاملQuality of boar spermatozoa from the sperm-peak portion of the ejaculate after simplified freezing in MiniFlatpacks compared to the remaining spermatozoa of the sperm-rich fraction.
Boar sperm viability post-thaw differs depending on the ejaculate fraction used, with spermatozoa present in the first 10 mL of the sperm-rich fraction (SRF) (portion 1, P1, sperm-peak portion) displaying the best cryosurvival in vitro compared with that of spermatozoa from the rest of the ejaculate (portion 2 of the SRF plus the post-spermatic fraction), even when using simplified freezing rou...
